Mediabox does not include an option to display a popup on mouseover. You would have to do this manually, eg: 

<a href="https://www.joomlacontenteditor.net/images/imagejpg" class="jcepopup" onmouseover="jcepopup.open(this.href);" data-mediabox="1" data-mediabox-width="640" data-mediabox-height="480">Image</a>
